小鼠双侧颈总动脉狭窄脑白质损伤模型的建立White Matter Damage in Mice with Bilateral Common Carotid Artery Stenosis

摘要(Abstract):
目的:建立双侧颈总动脉狭窄(BCAS)所致慢性低灌注脑白质损伤小鼠模型。方法:C57BL/6J小鼠42只,随机分为假手术组和模型组各21只,建立慢性低灌注脑白质损伤模型,假手术组不造成狭窄,利用7T超高场显微磁共振、髓鞘染色和电镜技术检测白质区病变,利用八臂迷宫行为学进行认知功能评判。结果:DTI相关参数显示,代表白质纤维完整性的部分各向异性在模型组的视束、内囊、胼胝体等白质区明显下降(P<0.05),代表脱髓鞘损伤的垂直扩散向量在胼胝体区明显升高(P<0.05),代表轴突损伤的轴向扩散向量在各白质区均未见明显变化;LFB染色和电镜亦证明白质区有明显脱髓鞘损伤。行为学试验证明模型组有明显的工作记忆错误(P<0.001),无明显参考记忆错误,且工作记忆错误和DTI参数所示脑白质损伤之间明显相关(P<0.05)。结论:小鼠BCAS 30 d可以造成选择性白质损伤,并检测到明显的工作记忆损害,两者之间存在相关性。
